The aim of the course:
The course aims to expose the students to microscopic analyses of wood and paper fibre, practical application of maceration process, micro slide making techniques, analysis of digital photos by the image analyze computer program, statistical evaluation of the data and presenting of results.

Upon successful completion of the course, the students will be able to :

Contents of the Course
Types of microscopes. Preparing of specimen and microslides. Microscopical analyses on fibre Type and its origin. Stains and their staining reactions. Determination of pulp type. Pulping process and the amount of bleaching according to staining reactions. Measuring of fiber size, Sheet strength and fiber size relations examination of cross section of paper. Examination of paper surface.

 Week 2Maceration of nonwood and wood plant sample
 Week 3Maceration of paper sample
 Week 4Preparation of temporary and permanent microslides from fiber samples
 Week 5Observations of microslides on the microscope: obtaining of black-white of coloured imges with polarised light filter
 Week 6Setting of DIGIMIZER image analyse computer program, testing of its properties and possibilities using in the micrographic study
 Week 7Measuring of dimension of softwood and/or hardwood fiber samples by DIGIMIZER image analyse computer program, Homework 1: studenf will prepare a rapor on measuring of fiber dimension for a given sample
 Week 8Analysis of fiber constituents of paper, recognation of mehanical pulp and chemical pulp fibers with pholoroglucine dye, Homework 2: studenf will prepare a rapor on determination of fiber source for a given paper sample
 Week 9Mid-term exam
 Week 10Preparation of standard iode based dye (indicator)
 Week 11Identification of pulp origin with iode based dyes
 Week 12Determination of springwood-summerwood and/or some cell ratios on the cross section of a plant sample, Homework 3: students will calculate of ratios of vessel, tracheid and paranchima cell in the plant tissue by the DIGIMIZER image analyse computer program
 Week 13Analysing of printed and unprinted paper surface by a light microscope
 Week 14Using of electon microscopy on observation of fiber surface, a presentation will be presented to the students
 Week 15Analysing of fotos taken scanned paper surface by DIGIMIZER image analyse computer program and determimation of dith and printed areas, Homework 4: printed paper samples will be given to student and will be wanted calculation of printed area of the paper
